Originally developed for studying the hormonal requirements of cultured cells, DMEM/Ham’s F12 is a widely used culture medium that serves as a base for the growth of various mammalian cell lines, such as MDCK cells, fibroblasts, or endothelial cells. It is a 1:1 mixture of DMEM (Dulbecco’s Modified Eagle Medium) and Ham’s F12, combining the nutrient richness of DMEM with the additional components of F12, including vitamins, trace elements, and amino acids.
